FORMULASI DAN EVALUASI MATRIKS TRANSDERMAL ATENOLOL
MENGGUNAKAN PROPILEN GLIKOL SEBAGAI ENHANCER SUBSTANCE
INTISARI
Atenolol merupakan obat golongan β-1 blocker selektif yang banyak
digunakan untuk mengobati hipertensi, angina pectoris, dan infark miokard.
Atenolol memiliki bioavailabilitas yang tidak cukup tinggi yaitu sekitar 50%,
sehingga atenolol ini dapat dibuat dalam sediaan transdermal. Sediaan transdermal
adalah suatu sistem dimana pengiriman bahan aktif dari obat terjadi melalui kulit.
Propilen glikol merupakan salah satu enhancer golongan glikol yang dapat
meningkatkan penetrasi, sehingga diharapkan atenolol dapat melalui kulit dan
dapat menembus lapisan stratum korneum. Penelitian ini bertujuan untuk
mengetahui pengaruh variasi kadar propilen glikol dalam sediaan matriks terhadap
sifat fisik matriks dan mengetahui kadar yang menghasilkan penetrasi optimal.
Formulasi matriks dilakukan dengan metode solvent casting dengan variasi kadar
propilen glikol 10%, 20%, dan 40%. Evaluasi yang dilakukan adalah uji sifat fisik
matriks transdermal meliputi uji organoleptis, uji keseragaman bobot, uji pH, uji
ketebalan, uji folding endurance, uji keseragaman zat aktif, serta uji permeasi dan
uji transport secara in vitro. Analisis hasil dilakukan dengan analisa statistik
dengan oneway ANOVA dilanjutkan dengan uji Tukey dan Independent Sample T
Test. Hasil penelitian menunjukkan bahwa propilen glikol memberikan perubahan
yang signifikan terhadap sifat fisik matriks transdermal atenolol dan semakin
tinggi jumlah propilen glikol maka semakin tinggi pula jumlah atenolol yang
tertransport.

ATENOLOL USING PROPYLENE GLYCOL AS ENHANCER SUBSTANCE
ABSTRACT
Atenolol is a drug class selective β-1 blockers are widely used to treat
hypertension, angina pectoris, and myocardial infarction. Atenolol has
bioavailability not sufficiently high which is about 50%, so that it can be made in
the atenolol transdermal preparations. A transdermal is a delivery system which
the active ingredient of the drug through the skin occurs. Propylene glycol is one
of the glycol group enhancer to increase penetration, so expect atenolol can be
through the skin and can penetrate the stratum corneum. This study aims to
determine the effect of variations in the levels of propylene glycol in the matrix
transdermal of the physical properties of the matrix and determine levels that
produce optimum penetration. Matrix formulation is performed by solvent casting
method with variations in levels of propylene glycol 10%, 20%, and 40%.
Evaluation is performed is test the physical properties of the matrix transdermal
include organoleptic test, weight uniformity test, pH test, test thickness, folding
endurance test, test uniformity of active substances, as well as the permeation test
and test transport in vitro. Analysis of the results performed by oneway ANOVA
followed by Tukey's test and Independent Sample T Test. The results showed that
the propylene glycol provide significant changes to the physical properties of the
matrix transdermal atenolol and the highest propylene glycol produce highest
percent of the transport atenolol.
