We report here the results of the structural analyses of this variant. Struc- The propositus was a healthy 43-year-old male. His hemato- logical data were as follows: PCV 0.48 1/1; MCV 95.6 fl; MCH 24.9 pg; and MCHC 26.0 g/dl. Alkali denaturation test showed 0.75% Hb F. No inclusion bodies were found in the red blood cells. Family studies showed that the mother and the two daughters of the propositus are also Hb Duan heterozygotes. Hb 12.5 g/dl; RBC 5.05 x 1OI2/1; Target cells were absent. Cellulose acetate membrane electrophoresis of the hemoglobin solution in TEB buffer, pH 8.6, showed an abnormal band between that of Hb A and Hb A2 (Fig. 1, Hb X) amounting t o 24.8% of the total hemoglobin content by densitometric measurement. A rather faint abnormal band (Hb Az-X), which moves more slowly than Hb Ap, was seen. It amounted to 1.6% of the total. The Hb A2 content of the propositus was 2.6%. Globin was prepared from the hemolysate by the acid-acetone method (1). Chain separation on cellulose acetate membrane electrophoresis (2) shoved it t o be a slowly-moving c1 chain variant. The abnormal cx chain was then isolated on a prepara- tive scale by the method of Clegg -- et al. (3) using a CM-cellulose (CM-23) chromatographic column.
