TOMM40 ‘523 variant and cognitive decline in older persons with APOE ε3/3 genotype
SUPPLEMENTAL METHODS
Tests for annual cognitive assessment
Uniform and structured cognitive testing was administered each year via a battery of 17 tests.
Five cognitive domains were assessed. Specifically, episodic memory was assessed using 7 tests,
which include immediate and delayed recall of story A from the logical memory subtest of the
Wechsler Memory Scale-Revised, immediate and delayed recall of the East Boston story, word
list memory, word list recall, and word list recognition. Semantic memory was assessed using 3
tests, including Boston naming, national adult reading and category fluency tests. Working
memory was assessed using 3 tests, including digit span forward, digit span backward and digit
ordering. Perceptual speed was assessed using 2 tests, including symbol digit modalities and
number comparison. Finally, visuospatial ability was assessed using 2 tests, including judgment
of line orientation and standard progressive matrices. Raw scores on each test were standardized
using the baseline mean and standard deviation of the entire cohorts. Individual standardized test
scores were averaged to obtain composite measures of global cognition and individual cognitive
domains. A higher score indicates better cognitive performance.
Figure e-1
Figure e-1 shows the histograms of the length (number of T residues) for each TOMM40 ‘523 allele among APOE ε3/3 carriers. The vertical dotted lines reflect the cut-offs used to determine the ‘523 categorical genotypes (S, L and VL).
