Cell Line: In vitro cultured primary hip-pocampal neurons
Method of validation: Immunofluorescence
Primary Antibody: STJ90311 Anti-Phospho-IκB-α (S32/S36)
Secondary Antibody: Donkey anti-rabbit AF568
Dilution ratio: 1:250, 1:1000
Protocol
Neurons were fixed with 2%PFA
Cells were washed 3 times for 5 minutes with PBS
Cells were permeabilised with 0.2% triton X100 for 5 minutes at RT
Cells were washed 3 times for 5 minutes with PBS
Blocking occured with 0.5% fish gelatin in PBS for 1 hour at 37ºC
The primary antibody was incubated overnight at 4ºC and then diluted to 1:250 or 1:1000 in 0.5% fish gelatin in PBS
Cells were washed 3 times for 5 minutes with PBS
The secondary antibody was incubated at RT for 1 hour and then diluted to 1:1000 in 0.5% fish gelatin in PBS
Cells were washed 3 times for 5 minutes with PBS
Cells were mounted in fluoroshield with DAPI moulding medium
