CD2, also known as LFA-2, is a member of the immunoglobulin gene family with an approximate molecular weight of 45 kD� It is expressed on thymocytes, peripheral T cells, dendritic epider-mal T cells, macrophages in the spleen, and NK cells� CD2 can as-sociate with the T cell receptor and participate in T cell activation through adhesion and signal transduction� CD2 binds to CD48, CD58, and CD59�

Clone: OX-34 Isotype: Mouse IgG2a, κThe OX-34 antibody blocks CD48 binding to CD2, partially blocks T cell activation, and inhibits the mixed lymphocyte reaction in vitro� The OX-34 antibody has also been used to deplete periph-eral T cells and prevent allograft rejection in vivo�

FOXP3 is a 50–55 kD transcription factor also known as Forkhead box protein P3, Scurfin, JM2, or IPEX� It is proposed that it is a master regulatory gene and more specific marker of T regula-tory cells than most cell surface markers (such as CD4 and CD25)� Transduced expression of FOXP3 in CD4+/CD25– cells has been shown to induce GITR, CD103, and CTLA4 and impart a T regula-tory cell phenotype� FOXP3 is mutated in X-linked autoimmunity-allergic dysregulation syndrome (XLAAD or IPEX) in humans and in “scurfy” mice� Overexpression of FOXP3 has been shown to lead to a hypoactive immune state, suggesting that this transcriptional factor is a central regulator of T cell activity�
