Transcript of Immunofluorescence Antibody Customer Review for Anti-β III Tubulin Antibody (STJ97123)
Result
“In primary hippocampal neurons STJ97123 detected the microtubule cytoskeleton. Antibody is specific, as no reac-tivity was detected in non-neuronal cells.”- P. Brites , Instituto de Biologia Molecular e Celular-IBMC.
Cell Line: In vitro cultured primary hip-pocampal neurons at DIV10
Method of validation: Immunofluorescence
Primary Antibody: STJ97123 Anti-β III Tubulin antibody
Secondary Antibody: Donkey anti-rabbit AF568
Dilution ratio: 1:250
Reference Antibody Mouse anti-β III Tubulin from
Millipore
Treatment of materials: At DIV10 Cells were fixed with 4% PFA in PBS
Protocol
Neurons were fixed with 4%PFA
Cells were washed 3 times for 5 minutes with PBS
Cells were permeabilised with 0.2% triton X100 for 5 minutes at RT
Cells were washed 3 times for 5 minutes with PBS
Blocking occured with 0.5% fish gelatin in PBS for 1 hour at 37ºC
The primary antibody was incubated overnight at 4ºC and then diluted to 1:500 in 0.5% fish gelatin in PBS
Cells were washed 3 times for 5 minutes with PBS
The secondary antibody was incubated at RT for 1 hour and then diluted to 1:1000 in 0.5% fish gelatin in PBS
Cells were washed 3 times for 5 minutes with PBS
Cells were mounted in fluoroshield with DAPI moulding medium
