IgG LCK LYN FYN HCK c-S R C phospho -SFK β-actin Supplemental Figure 1 Supplemental Figure 1. Lyn is the predominant activated SFK in AML primary cells. Whole cell lysate of primary AML FAB M5 cells was subjected to immunoprecipitation with control IgG antibody, or antibodies directed against LCK, LYN, FYN, HCK, or c-SRC. Following immunoprecipitation of the different SFKs, the immunodepleted lysates were subjected to immunoblotting with anti-phospho-SFK antibody (- phospho-SRC Y418). The blots of immunodepleted lysates were stripped and reprobed with anti--actin to demonstrate equal protein input. Immunodepletion of LYN resulted in removal of the phospho-SFK immunoreactive band. The experiment was performed twice with similar results.
