Goals of work

Investigate the involvement of the adipokines eotaxin-3,(MIP-1β)macrophage inflammatory protein-1β , and( MCP-4) monocyte chemotactic protein-4 in obesity and related comorbidities and the modification of their circulating levels after bariatric surgery.

Subjects & Method

Subjects:• 80 obese patients(25 male -55 female in age

between 11-43)in list of bariatric surgery (laparoscopic sleeve gastrectomy -LSG) were examined at the beginning of the research and 10 months after LSG, while normal-weight controls were examined only once at beginning of the research.

• 20 control (9 male -11 female ,age between 12-48)

Subjects & MethodMethod: Anthropometric, clinical and laboratory

parameters Measurements of plasma levels of eotaxin-3,

MIP-1β, MCP-4, leptin and adiponectin :• Blood was collected then plasma was separated

using centrifuge .• Leptin and adiponectin were measured by an

enzyme-immunoassorbent assay (ELISA) .• Eotaxin-3, MIP-1β, MCP-4 were measured by

(ECL) electrochemiluminescence immunoassay.

• Electrochemiluminescence permits the detection and measurement of a biological or chemical substance within a sample.

Subjects & Method

Ruthenium and TPA are oxidized at the surface of the electrode when voltage is applied. The TPA loses a proton, which reduces the ruthenium to an excited state, causing light to be emitted. Multiple excitation/emission cycles amplifies the light emitted and increase sensitivity. The emitted light is measured to determine concentration of analytes in sample.

Subjects & Method Instrumental analysis: Intima-media

thickness measurementCarotid arteries ( right and left ) were examined by

high-resolution B-mode ultrasonography using ultrasound device.

Result

A. all the anthropometric , clinical and metabolic characteristics are higher in the obese group before-surgery in comparative with the control except (TC, LDL-C, HDL.C-TG and ApoB) .

after surgery , all the anthropometric , clinical and metabolic characteristics are highly decreased except HDL.C increased and TC, LDL-C has no changes.

obesity because their decreasing with the decrease of BMI

• it is play indirect role in obesity related diseases by affecting the other chemokines (eotaxin -3 and MIP- 1β)

• MIP-1β and eotaxin-3 are Play an important role in diseases associated with obesity, such as inflammation and atherosclerosis and heart and blood vessels disorders .
